    Assessing the organic fraction of municipal solid wastes for the production of lactic acid

    With an estimated yearly production of about 140 Mt in the EU, conventionally, the organic fraction of municipal solid wastes (OFMSW) has been disposed in landfills with negative environmental effects. Nonetheless, the chemical composition of this residue make it a substrate with great bioconversion potential. In this study, OFMSW from Spanish municipal treatment plants, were evaluated for the production of LA. Samples were identified according to the sorting mechanisms employed for their collection in: (A) separately collected, (B) non-separately collected and (C) separately collected+paper/cardboard. Enzymatic hydrolysis was used to produce hydrolysates A, B and C accordingly. Hydrolysate A showed the highest total sugars and glucose content with values of 70 and 55 g·L−1, respectively. Following the characterisation, a screening showed that growth of B. coagulans was possible in all three hydrolysates. Furthermore, lab scale fermentations showed that LA final concentrations could reach around 60 g·L−1, with yields from total sugars of above 0.60 g·g−1. A technical scale fermentation of the hydrolysate A resulted in a final LA concentration of 60.7 g·L−1, a yield of 0.71 g·g−1 with a productivity of 2.68 g·L−1·h−1. Overall, it was estimated that 0.23 g of LA could be produced from one g of dry OFMSW.

    Rapid determination of lime requirement by mid-infrared spectroscopy: A promising approach for precision agriculture

    Mid-infrared spectroscopy (MIRS) has proven to be a cost-effective, high throughput measurement technique for soil analysis. After multivariate calibration mid-infrared spectra can be used to predict various soil properties, some of which are related to lime requirement (LR). The objective of this study was to test the performance of MIRS for recommending variable rate liming on typical Central European soils in view of precision agriculture applications. In Germany, LR of arable topsoils is commonly derived from the parameters organic matter content (SOM), clay content, and soil pH (CaCl2) as recommended by the Association of German Agricultural Analytical and Research Institutes (VDLUFA). We analysed a total of 458 samples from six locations across Germany, which all revealed large within-field soil heterogeneity. Calcareous topsoils were observed at some positions of three locations (79 samples). To exclude such samples from LR determination, peak height at 2513 cm−1 of the MIR spectrum was used for identification. Spectra-based identification was accurate for carbonate contents > 0.5%. Subsequent LR derivation (LRSPP) from MIRS-PLSR predictions of SOM, clay, and pH (CaCl2) for non-calcareous soil samples using the VDLUFA look-up tables was successful for all locations (R2 = 0.54–0.82; RMSE = 857–1414 kg CaO ha−1). Alternatively, we tested direct LR prediction (LRDP) by MIRS-PLSR and also achieved satisfactory performance (R2 = 0.52–0.77; RMSE = 811–1420 kg CaO ha−1; RPD = 1.44–2.08). Further improvement was achieved by refining the VDLUFA tables towards a stepless algorithm. It can be concluded that MIRS provides a promising approach for precise LR estimation on heterogeneous arable fields. Large sample numbers can be processed with low effort which is an essential prerequisite for variable rate liming in precision agriculture. © 2019 The Authors.     Relevance of interactions between starch-based coatings and plum fruit surfaces: A physical-chemical analysis

    In order to extend the shelf life of the fruit, improve appearance, and to keep all nutrition properties of the plum from diminishing, edible coatings comprised of wheat starch and wheat starch–whey protein isolate (in ratio 80/20) were created. Stand-alone films were produced to assess properties which helped to understand the phenomena occurring on the surface level of coated plums. The properties of coatings based on starch are similar to starch coatings containing oil because the natural epicuticular wax layer of plums merges with coating materials. Adding oil doubled the contact angle value and the dispersive component of the surface tension. The workings of adhesion and cohesion, spreading coefficient, water absorption, water content, and solubility in water of the films decreased. Similar processes were observed on the fruits’ surface. In appearance, the coating process is similar to polishing the plum surface for removing crystalline wax. The color parameters of coated fruits did not significantly change. Newly formed bonds or interactions established between starch, whey proteins, water, glycerol, and oil are displayed by Fourier transform infrared (FTIR) analysis. This work revealed how the interactions between the epicuticular wax on the fruit’s surface and the hydrocolloid-based coatings affect the efficiency of the coatings. © 2019 by the authors.     Metaproteome analysis reveals that syntrophy, competition, and phage-host interaction shape microbial communities in biogas plants

    Background: In biogas plants, complex microbial communities produce methane and carbon dioxide by anaerobic digestion of biomass. For the characterization of the microbial functional networks, samples of 11 reactors were analyzed using a high-resolution metaproteomics pipeline. Results: Examined methanogenesis archaeal communities were either mixotrophic or strictly hydrogenotrophic in syntrophy with bacterial acetate oxidizers. Mapping of identified metaproteins with process steps described by the Anaerobic Digestion Model 1 confirmed its main assumptions and also proposed some extensions such as syntrophic acetate oxidation or fermentation of alcohols. Results indicate that the microbial communities were shaped by syntrophy as well as competition and phage-host interactions causing cell lysis. For the families Bacillaceae, Enterobacteriaceae, and Clostridiaceae, the number of phages exceeded up to 20-fold the number of host cells. Conclusion: Phage-induced cell lysis might slow down the conversion of substrates to biogas, though, it could support the growth of auxotrophic microbes by cycling of nutrients. © 2019 The Author(s).

    Process disturbances in agricultural biogas production—causes, mechanisms and effects on the biogas microbiome: A review

    Disturbances of the anaerobic digestion process reduce the economic and environmental performance of biogas systems. A better understanding of the highly complex process is of crucial importance in order to avoid disturbances. This review defines process disturbances as significant changes in the functionality within the microbial community leading to unacceptable and severe decreases in biogas production and requiring an active counteraction to be overcome. The main types of process disturbances in agricultural biogas production are classified as unfavorable process temperatures, fluctuations in the availability of macro- and micronutrients (feedstock variability), overload of the microbial degradation potential, process-related accumulation of inhibiting metabolites such as hydrogen (H 2 ), ammonium/ammonia (NH 4 + /NH 3 ) or hydrogen sulphide (H 2 S) and inhibition by other organic and inorganic toxicants. Causes, mechanisms and effects on the biogas microbiome are discussed. The need for a knowledge-based microbiome management to ensure a stable and efficient production of biogas with low susceptibility to disturbances is derived and an outlook on potential future process monitoring and control by means of microbial indicators is provided.

    Reducing conditions favor magnetosome production in magnetospirillum magneticum AMB-1

    Magnetotactic bacteria (MTB) are a heterogeneous group of Gram-negative prokaryotes, which all produce special magnetic organelles called magnetosomes. The magnetosome consists of a magnetic nanoparticle, either magnetite (Fe3O4) or greigite (Fe3S4), embedded in a membrane, which renders the systems colloidaly stable, a desirable property for biotechnological applications. Although these bacteria are able to regulate the formation of magnetosomes through a biologically-controlled mechanism, the environment in general and the physico-chemical conditions surrounding the cells in particular also influence biomineralization. This work thus aims at understanding how such external conditions, in particular the extracellular oxidation reduction potential, influence magnetite formation in the strain Magnetospirillum magneticum AMB-1. Controlled cultivation of the microorganisms was performed at different redox potential in a bioreactor and the formation of magnetosomes was assessed by microscopic and spectroscopic techniques. Our results show that the formation of magnetosomes is inhibited at the highest potential tested (0 mV), whereas biomineralization is facilitated under reduced conditions (-500 mV). This result improves the understanding of the biomineralization process in MTB and provides useful information in sight of a large scale production of magnetosomes for different applications. © 2019 Olszewska-Widdrat, Schiro, Reichel and Faivre.

    Respiratory patterns of European pear (Pyrus communis L. ‘Conference’) throughout pre- and post-harvest fruit development

    Information on the developmental stage of pear pre-harvest and in shelf-life is crucial to determine the optimum timing of harvest, post-harvest treatment, and time of consumption ensuring high eating quality. In the present study, CO2 emission and fruit quality of European pear (Pyrus communis L.) ‘Conference’ were analysed pre- and post-harvest with emphasis on shelf life for three years. Additionally, cytochrome and cyanide-resistant O2 consumption were analysed in the last year of experiments. The respiration rate of pear showed typical climacteric rise of CO2 emission in two years only, despite daily measurements. However, in each year the fruit quality in shelf life was closely linked to harvest date suggesting climacteric fruit response. Thus, the developmental stage of ‘Conference’ pear should be analysed by additional methods. Particularly, the cytochrome and cyanide-resistant O2 consumption showed an encouraging potential to obtain data on characteristic respiratory patterns. © 2019

    Production and purification of L-lactic acid in lab and pilot scales using sweet sorghum juice

    Sweet sorghum juice (SSJ) was evaluated as fermentation substrate for the production of l-lactic acid. A thermophilic Bacillus coagulans isolate was selected for batch fermentations without the use of additional nutrients. The first batch of SSJ (Batch A) resulted on higher lactic acid concentration, yield and productivity with values of 78.75 g·L−1, 0.78 g·g−1 and 1.77 g·L−1 h−1, respectively. Similar results were obtained when the process was transferred into the pilot scale (50 L), with corresponding values of 73 g·L−1, 0.70 g·g−1 and 1.47 g·L−1 h−1. A complete downstream process scheme was developed in order to separate lactic acid from the fermentation components. Coarse and ultra-filtration were employed as preliminary separation steps. Mono- and bipolar electrodialysis, followed by chromatography and vacuum evaporation were subsequently carried out leading to a solution containing 905.8 g·L−1 lactic acid, with an optical purity of 98.9%. The results of this study highlight the importance of the downstream process with respect to using SSJ for lactic acid production. The proposed downstream process constitutes a more environmentally benign approach to conventional precipitation methods.

    N 2 O emissions and NO 3 − leaching from two contrasting regions in Austria and influence of soil, crops and climate: a modelling approach

    National emission inventories for UN FCCC reporting estimate regional soil nitrous oxide (N 2 O) fluxes by considering the amount of N input as the only influencing factor for N 2 O emissions. Our aim was to deepen the understanding of N 2 O fluxes from agricultural soils, including region specific soil and climate properties into the estimation of emission to find targeted mitigation measures for the reduction of nitrogen losses and GHG emissions. Within this project, N 2 O emissions and nitrate (NO 3 − ) leaching were modelled under spatially distinct environmental conditions in two agricultural regions in Austria taking into account region specific soil and climatic properties, management practices and crop rotations. The LandscapeDNDC ecosystem model was used to calculate N 2 O emissions and NO 3 − leaching reflecting different types of vegetation, management operations and crop rotations. In addition, N input and N fluxes were assessed and N 2 O emissions were calculated. This approach allowed identifying hot spots of N 2 O emissions. Results show that certain combinations of soil type, weather conditions, crop and management can lead to high emissions. Mean values ranged from 0.15 to 1.29 kg N 2 O–N ha −1  year −1 (Marchfeld) and 0.26 to 0.52 kg N 2 O–N ha −1  year −1 (Grieskirchen). Nitrate leaching, which strongly dominated N-losses, often reacted opposite to N 2 O emissions. Larger quantities of NO 3 − were lost during years of higher precipitation, especially if winter barley was cultivated on sandy soils. Taking into account the detected hot spots of N 2 O emissions and NO 3 − leaching most efficient measures can be addressed to mitigate environmental impacts while maximising crop production. © 2018, The Author(s).

    Validation study for measuring absorption and reduced scattering coefficients by means of laser-induced backscattering imaging

    Decoupling of optical properties appears challenging, but vital to get better insight of the relationship between light and fruit attributes. In this study, nine solid phantoms capturing the ranges of absorption (μa) and reduced scattering (μs’) coefficients in fruit were analysed non-destructively using laser-induced backscattering imaging (LLBI) at 1060 nm. Data analysis of LLBI was carried out on the diffuse reflectance, attenuation profile obtained by means of Farrell's diffusion theory either calculating μa [cm−1] and μs’ [cm−1] in one fitting step or fitting only one optical variable and providing the other one from a destructive analysis. The nondestructive approach was approved when calculating one unknown coefficient non-destructively, while no ability of the method was found to analysis both, μa and μs’, non-destructively. Setting μs’ according to destructive photon density wave (PDW) spectroscopy and fitting μa resulted in root mean square error (rmse) of 18.7% in comparison to fitting μs’ resulting in rmse of 2.6%, pointing to decreased measuring uncertainty, when the highly variable μa was known. The approach was tested on European pear, utilizing destructive PDW spectroscopy for setting one variable, while LLBI was applied for calculating the remaining coefficient. Results indicated that the optical properties of pear obtained from PDW spectroscopy as well as LLBI changed concurrently in correspondence to water content mainly. A destructive batch-wise analysis of μs’ and online analysis of μa may be considered in future developments for improved fruit sorting results, when considering fruit with high variability of μs’. © 2019 The Authors
