    Real-time image processing for label-free enrichment of Actinobacteria cultivated in picolitre droplets

    The majority of today's antimicrobial therapeutics is derived from secondary metabolites produced by Actinobacteria. While it is generally assumed that less than 1% of Actinobacteria species from soil habitats have been cultivated so far, classic screening approaches fail to supply new substances, often due to limited throughput and frequent rediscovery of already known strains. To overcome these restrictions, we implement high-throughput cultivation of soil-derived Actinobacteria in microfluidic pL-droplets by generating more than 600000 pure cultures per hour from a spore suspension that can subsequently be incubated for days to weeks. Moreover, we introduce triggered imaging with real-time image-based droplet classification as a novel universal method for pL-droplet sorting. Growth-dependent droplet sorting at frequencies above 100 Hz is performed for label-free enrichment and extraction of microcultures. The combination of both cultivation of Actinobacteria in pL-droplets and real-time detection of growing Actinobacteria has great potential in screening for yet unknown species as well as their undiscovered natural products.

    Der Ansatz von Precision Horticulture im Obstbau lehnt sich an das aus dem Ackerbau stammende Konzept der Präzisionslandwirtschaft bzw. der teilflächenspezifischen Bewirtschaftung an. Hierbei sollen präzise an das individuelle Gehölzwachstum angepasste Pflegemaßnahmen die bislang praktizierte einheitliche Behandlung aller Bäume in einer Anlage ablösen. Voraussetzungen hierfür sind u. a. Bodenkarten und Informationen zum Pflanzenwachstum. Das Ziel ist es, den informationsgestützten Obstbau voranzutreiben und durch ein räumlich und zeitlich differenziertes Management eine effizientere und nachhaltigere Bewirtschaftung zu erreichen.

    When humans and machines collaborate: Cross-lingual Label Editing in Wikidata

    The quality and maintainability of a knowledge graph are determined by the process in which it is created. There are different approaches to such processes; extraction or conversion of available data in the web (automated extraction of knowledge such as DBpedia from Wikipedia), community-created knowledge graphs, often by a group of experts, and hybrid approaches where humans maintain the knowledge graph alongside bots. We focus in this work on the hybrid approach of human edited knowledge graphs supported by automated tools. In particular, we analyse the editing of natural language data, i.e. labels. Labels are the entry point for humans to understand the information, and therefore need to be carefully maintained. We take a step toward the understanding of collaborative editing of humans and automated tools across languages in a knowledge graph. We use Wikidata as it has a large and active community of humans and bots working together covering over 300 languages. In this work, we analyse the different editor groups and how they interact with the different language data to understand the provenance of the current label data.
